ImmunoHistoMount (IHM) ™ Medium, A Permanent Aqueous Mounting Medium

Catalog number: AR-6503-01 (30 ml), AR-6503-02 (100 ml), AR-6503-03 (250 ml), AR-6503-05 (1000 ml).

Description: Immunohistomounting medium (IHM) is a permanent aqueous mounting medium designed for permanent mounting of tissue sections and cell smears with peroxidase and alkaline phosphatase chromogens that can not be dehydrated with organic solvents.  This mounting medium preserves Fast red, Aminoethylycarbazole (AEC), BCIP/NBT, BCIP/INT chromogens and is also compatible with counterstain like Hematoxylin and Nuclear fast red (NFR). It is also suitable for chromogens like DAB and DAB with nickel and cobalt. It is not compatible with H and E staining.

Intended Use: Mounting of immunohisto slides that cannot be dehydrated with organic solvents.

Reagent: Ready to use mounting medium

Storage: This product is stored at room temp, however for long term storage 2-8oC is recommended. DO NOT FREEZE.

Procedure:

  1. Bring the vial to room temperature.
  2. Rinse slide to be mounted with DISTILLED OR DEIONIZED WATER, touch the edges of slide on a paper towel to remove excess water. Place slides on a flat surface.
  3. Turn the vial upside down and open the dropper to remove any air bubbles.
  4. Apply 3-4 drops of mounting medium directly on top of the specimen and spread out evenly by tilting the slide back and forth or spread evenly with a 0.2 ml plastic pipette tip making sure the tissue is not touched. Excess medium can be removed by touching the edges of slide against a paper towel.
  5. Let stand at room temperature for about 1 to 2 hours. Or the slides can be heated at 37-40oC for 40-60 minutes. The slides are ready for visualization under a microscope.

There are two ways to look slide under oil immersion lens.

1.      For oil immersion lens, a drop of oil can be placed directly on dried ImmunoHistoMount mounting medium for visualization.

The oil can be removed by wiping with Kimwipes          OR

2.      Apply 1-2 drops of any organic mounting medium on top of dried ImmunoHistoMount mounting medium; which has protected the specimen, cover slide with coverslip; the slide is ready for visualization.

 

 Removal of mounting medium: If the coverslip has not been applied, soak the slide in warm (37oC) water for 5-10 minutes until the mounting medium is dissolved. Rinse slide with warm water to remove all mounting medium. The slide can be remounted again.

MSDS: This product contains 0.05 % sodium azide as a preservative, appropriate care should be taken in handling. National Institute of Occupational Safety and Health has warning that sodium azide can react with lead, copper, brass or solder in the plumbing system and forms hydrazoic acid in acidic condition. Discard with copious amount of water. Avoid skin and eye contact with all laboratory products. Use appropriate lab. gear, lab coat , gloves and  safety glasses. Do not ingest any lab. products.
