INSECTOGENE
Transfection Reagent Specific for Insect Cells
The transfection reagent
INSECTOGENE is an aqueous formulation of positively charged lipids
with a concentration of 1 mg/ml. It is especially designed for optimum
liposome-mediated transfection of insect cells. The efficiency is much
higher than transfection of insect cells with Ca-phosphate, DEAE-Dextrane
and other competing reagents.
Insect cells play an important role for the baculovirus expression vector
system (BEVS), which was introduced in 1983 by Smith et al.
1. BEVS has become one of the most
powerful eukaryotic vector systems for recombinant protein expression.
Baculoviruses are not pathogenic to mammals and plants, they have a very
restricted host range mostly limited to invertebrate species. Pathogenic
viruses never transform insect cells; therefore, minimal containment
conditions are required. Insect cells can be easily and reproducibly scaled
up for large-scale production of recombinant proteins. For example, cell
lines derived from Spodoptera frugiperda can easily be cultivated as
suspension cultures, which is the supposition for large-scale production in
bioreactors. The most used cell lines for BES applications are Spodoptera
frugiperda (Sf9, Sf-21) and Trichoplusia ni. (e.g. Tn-368)

Comparison of Transfection Efficiencies
Transfection assays were performed with optimized protocols using
pBacMamßGal or similar genes. Efficiencies were assayed by counting of
plaques after X-Gal-staining.
