METAFECTENE®
The Highly Efficient RMA-based Transfection Reagent for Mammalian Cells
Product description
The high efficiency of METAFECTENE® makes it a valuable tool for gene delivery or gene transfer experiments.
METAFECTENE® is provided as a ready-to-use solution. Storage at 4°C for more than a year does not affect its transfection efficiency. It was also shown to be stable for weeks at 40°C. In contrast to most other transfection reagents METAFECTENE® shows no serum inhibition, which makes it the reagent of choice for transfecting sensitive cell lines. Depending on the cell line, 1 ml METAFECTENE® is sufficient for 50-200 transfections on 35 mm dishes (6-well plates) or 15 to 70 transfections on 60 mm dishes.
While METAFECTENE® is suitable for a broad spectrum of applications, one of its best-known properties is its compatibility with high-throughput applications such as high-throughput screening (HTS). Other transfection methods either are not HTS compatible or need high amounts of DNA and reagent.
Highlights of METAFECTENE®
- Extremly fast and easy procedure
- For use in high-throughput systems
- No serum inhibition
- Broad cell line range
- Best results with siRNA applications
- High efficiency by use of very low amounts of DNA
- Broad peak performance
- Excellent reproducibility
- Low toxicity
- Best value for moneyZellen
RMA-Technology
The unique properties of the reagent METAFECTENE® are based on RMA-Technology ("Repulsive Membrane Acidolysis"), which ensures high transfection efficiency with low amounts of DNA and reagent. The combination, with its extremely simple instructions for use, delivers an outstanding price/performance ratio, an important consideration for both standard transfection procedures and especially for high-throughput assays.
The specifically designed molecular structure of METAFECTENE® and METAFECTENE® PRO provides for the following processes:
Highlights of METAFECTENE®
- Easy entry of genetic material into cells by condensing DNA into compact structures
- Endosome buffering for osmotic destruction of the endosome membrane (proton sponge properties)
- Destabilisation of the DNA-coating lipid membrane by repulsive electrostatic forces in weakly acidic environment of late endosomes
Extremely fast and easy procedure with METAFECTENE®
Comparison of METAFECTENE® with 5 other well known transfection reagents
Cells were transfected in triplicate with 0.1µg pND2Luc plasmid on 96 well plates. Luciferase activities were quantitated in a Promega luciferase assay by using a luminometer and are given as relative values of maximum transfection efficiencies (100%) for reagents with each cell line.
