Buffer The antibodies are supplied with 1% BSA in PBS with 0.05% azide, pH 7.4. The Prediluted antibody is supplied in our Universal antibody dilution buffer (AR-6526) green in color. Description: Sialosyl Tn antigen also known as CD175s is expressed in the majority of colon, gastric, breast and lung carcinoma. The specificity of this antibody is similar to B 72.3 antibody. The sialosyl Tn antigen has a limited expression to being absent in normal colon, pancreas, stomach, ovary, and prostate. This antigen is detected in several adenocarcinomas. It appears to be useful marker for poorly differentiated colon adenocarcinomas. This antibody stains colon carcinomas and ovarian mucinous carcinomas producing apical cytoplasmic, apical membrane or luminal staining patterns. It is negative with majority of adult tissue sections.
Intended Use: Immunohistochemistry (IHC) and Immunocytchemistry (ICC) Immunofluorescence (IF), Flow cytometry (not tested at our lab). Storage: 2-8°C Clone: STn 219 Isotype: IgG1/κ Epitope: NeuAc α GalNacOSer/Thr Molecular weight of antigen: several HM weight glycoproteins. Immunogen: .Ovine submaxillary mucin (OSM) Species reactivity: Human, others not tested. Cellular Localization: Cytoplasmic, apical membrane, luminal Recommended positive control: Human gastrointestinal tumor, prostate, ovary ca. Application: IHC, ICC (frozen or formalin–fixed paraffin-embedded (FFPE) tissue sections, cell smears. For IHC dilute conc. antibodies 1:50-1:100, use streptavidin~biotin system or polymer system, incubate 30 minutes at room temperature. Prediluted antibody is ready to be used for IHC. IF, FITC~conjugated antibodies 10-20 µg/ml (1:10-1:20), incubate for 2 hours in the dark at RT or it can also be incubated overnight at 4ºC. Flow: FITC~conjugated antibodies, 0.2-1.0 µg/0.1 ml (1:200-1:1,000) (Not tested in our lab). The optimum dilution should be determined by the individual lab. General References 1. Gold, DV et. al.
