Tetramethylbenizidine MEMBRANE (TMBM) (Ready-to-use) Peroxidase Chromogen Substrate for Western, Northern, Southern, Slot or Dot Blots.(One component) Catalog number: AR-8223-01 50 ml AR-8223-02 100 ml AR-8223-04 500 ml Description: This 3,3'5,5’- tetramethylbenzidine (TMB) is a insoluble chromogen substrate for horseradish peroxidase detection systems for Membrane (for Western, Northern, Southern, Slot or Dot Blots). This is ready to use one component system and gives aqua marine bands with peroxidase. This TMB solution contains TMB, 1.13 mMol L-1, Hydrogen Peroxide substrate, 1% DMSO, acetate buffer, pH 4.9 and non-toxic proprietary stabilizers. Intended Use: Western. Northern, Southern, slot and slot or dot blots. It s ready to use and does not require any preparation. Storage: 2-8°C Application: 1. After the peroxidase reaction, wash membrane thoroughly with PBS or Tris-HCl saline containing 0.1 % Tween 20. DO NOT ADD THIMEROSAL, azide or phenolic compounds as preservative in any buffers. 2. Cover membrane with TMBM solution and incubate with gentle rocking for 5-30 minutes till aqua blue color bands appear. (If bands appear immediately; the concentration of peroxidase labeled probe is too high and should be further diluted). 3. Stop reaction by washing thoroughly in distilled or deionized water. DO NOT WASH WITH BUFFER OR TAP WATER. DO NOT INTRODUCE ANY ACID AT THIS STAGE, THE BANDS WILL TURN YELLOW.
The optimum time of incubation for TMB should be determined by the individual lab. ANOID EXPOSURE TO LIGHT Discard if the solution is blue or turbid.
The reaction mixture can be discarded according to city, county, state, province or country’s regulations.
